Summary: The human beta-globin gene cluster located on chromosome 11 spans about 45 kb and includes five functional genes and one pseudogene. The order of genes from 5' to 3' is: epsilon, G-gamma, A-gamma, beta 1 pseudogene, delta, and beta, or HBE1, HBG2, HBG1, HBBP1, HBD and HBB, respectively. These beta-like genes have the same overall exonic structure, leading to the conclusion that they are derived from one ancestral gene. The epsilon- and the two gamma-globin genes are expressed in early embryonic and fetal erythroid tissues, respectively, while the delta- and beta-globin genes are expressed in adult life. Developmental expression of these genes is controlled by a locus control region (LCR) that exists upstream of the HBE1 gene, and it includes five major DNase I hypersensitive sites and several enhancer regions. Two epsilon chains combine with two zeta chains to constitute the embryonic hemoglobin Hb Gower I, while two epsilon chains together with two alpha chains form the embryonic Hb Gower II. Two gamma chains combine with two alpha chains to make fetal hemoglobin HbF. In addition, two alpha chains plus two beta chains constitute HbA, which in normal adult life comprises about 97% of total hemoglobin. Mutations in either the gene cluster region or LCR can result in beta-thalassemias and beta-hemoglobinopathies. Sickle cell anemia and beta-thalassemia result from mutation and absence, respectively, of the beta-globin chain. [provided by RefSeq, May 2017].
