In RNA-Seq, the presence of highly expressed transcripts which may have limited biological relevance, such as ribosomal RNAs (rRNA), can overshadow the identification of more valuable low-abundance transcripts. This issue is particularly pronounced when confronted with sample types lacking accessible rRNA depletion solutions.

Addressing this issue custom riboPOOLs offer tailored solutions for eliminating unwanted rRNAs and other abundant RNAs from any organism. By designing complex pools of biotinylated DNA probes, specifically designed to hybridize and remove ribosomal RNAs (rRNA) from RNA samples, we can create riboPOOLs for:

  • Any species or mixture of RNA (e.g., seawater samples)

  • High quality to strongly degraded RNA (i.e., FFPE samples)

  • Standard RNA-Seq or Ribosome Profiling (Ribo-Seq) samples

"We are doing a lot of ribosome profiling experiments - mainly using mouse tissues and cell lines - and were in need of a ribosomal RNA depletion tool that was robust, economical, and adaptable. Initially, we asked siTOOLs Biotech to design specific antisense probes against the most abundant rRNA contaminants that we tended to see in our ribosome profiling data, and that severely reduced the amount of usable reads.
