E3 Ubiquitin Ligase siRNA Library

Potent & Specific Gene Knockdown for E3 Ligases

Gene silencing of E3 ubiquitin ligases

The E3 Ligase siPOOL Library contains siRNA pools designed to target E3 ubiquitin ligases involved in substrate recognition, protein ubiquitination and proteasomal degradation. These enzymes regulate essential processes including protein homeostasis, cell-cycle progression, DNA-damage responses, immune signalling and apoptosis.

Each E3 ligase gene is targeted using a complex pool of 30 carefully designed siRNAs. By combining broad transcript coverage with low concentrations of the individual siRNAs, siPOOL technology supports robust and reproducible gene knockdown while minimizing sequence-specific off-target effects—making the library an effective solution for focused and high-throughput RNAi screening.

E3 Ubiquitin Ligase siPOOL Library knockdown efficiency

Our E3 Ligase siPOOL Library delivers reliable gene silencing across a broad range of E3 ubiquitin ligase targets.

More than 91% of the tested E3 ligase siPOOLs reduced target-gene expression by at least 70% (30 of 33). In addition,76 % achieved at least 80% knockdown (Fig. 1).

The E3 Ligase siPOOL Library is ideal for:

  • High-throughput and focused RNAi screening.
  • Investigating ubiquitin signalling and protein-degradation pathways.
  • Identifying and validating E3 ligase functions and substrates.
  • Target discovery for PROTACs, molecular glues and other targeted protein-degradation approaches.
  • Studying E3 ligase involvement in cancer, neurodegenerative disorders, immune diseases and other pathologies.

E3 Ligase siPOOL Library features and benefits

  • Reduced off-target effects: Each of the 30 siRNAs is used at picomolar concentrations, limiting sequence-specific off-target activity.
  • Broad transcript coverage: The siRNAs are distributed across the annotated transcripts of each E3 ligase gene, reducing the risk of individual isoforms escaping silencing.
  • Robust and reliable knockdown: Combining 30 siRNAs reduces dependence on the performance of any single sequence and supports reproducible results.
  • Flexible, screen-ready formats: Available in customizable 96- and 384-well plate layouts with your preferred positive and negative controls.
  • e and negative controls.
